Studies on electron beam induced DNA damage and repair kinetics in lymphocytes by alkaline comet assay
Background: Exposure to ionizing radiation is known to induce oxidative stress followed by damage to critical biomolecules like lipids, proteins and DNA through radiolysis of cellular water. Since radiation has been widely used as an important tool in therapy of cancer, the detailed investigation regarding the DNA damage and repair kinetics would help to predict the radiation sensitivity of cel...
متن کاملRepair of ionizing radiation induced DNA damage in human lymphocytes.
Phytohemagglutinin stimulated human lymphocytes exhibit a 20 fold increase in DNA repair synthesis following ionizing radiation damage compared to the level of repair in unstimulated cells. The peak of repair synthesis coincides with that for DNA replication. Stimulated lymphocytes provide a relatively simple assay for ionizing radiation repair defects.
متن کاملDNA repair synthesis in subpopulations of human lymphocytes.
Human peripheral blood lymphocytes enriched in T or B cells were exposed to ultraviolet irradiation, nitrogen mustard or methylmethane sulphonate and investigated regarding their capacity for DNA repair synthesis. The DNA repair synthesis, measured as [3H] thymidine incorporated, was determined by autoradiography as unscheduled DNA synthesis (UDS). No systematic difference could be found in the...
متن کاملUltraviolet-induced DNA excision repair in human B and T lymphocytes. III. Repair in lymphocytes from chronic lymphocytic leukaemia.
There have been conflicting reports about the capacity of lymphocytes from individuals with chronic lymphocytic leukaemia (CLL) to undertake u.v.-induced DNA repair. We have examined repair in CLL cells and in control and age-matched purified B and T lymphocytes by a technique independent of incorporation of radioactive precursor, i.e. by the recovery of normal sedimentation behaviour of nucleo...
متن کاملInduction of the DNA repair enzyme uracil-DNA glycosylase in stimulated human lymphocytes.
The capacity of human cells to modulate the synthesis of DNA repair enzymes has been investigated by measuring the induction of the uracil-DNA glycosylase during lymphocyte stimulation. Treatment of peripheral lymphocytes with phytohemagglutinin increased glycosylase activity 10-fold. Glycosylase stimulation was coordinate with the activation of DNA synthesis and DNA polymerase activity. Two ch...
